ABSTRACT. Mating pair formation in Paramecium primaurelia was shown to be inhibited by incubating mating-competent mating type (mt) I and mt U cells with Limulus polyphemus agglutinin (LPA) or wheat germ agglutinin (WGA). Preincubation of LPA and WGA with their specific binding-monosaccharides, N-acetylneuraminic acid (NeuAc) and N-acetylglucosamine (GlcNAc), respectively, prevented the lectin effect on pair formation. Addition either of NeuAc or GlcNAc resulted in a reversal of cell pairing inhibition by LPA or WGA, respectively. Both NeuAc and GlcNAc monosaccharides inhibited pair formation when their concentration exceeded a threshold value. The pattern of the relative distribution of NeuAc and GlcNAc molecules on the cell surface was analyzed using fluorescence resonance energy transfer techniques combined with imaging systems. Mt I1 cells showed a high lectin-binding site density localized just on the surface region engaged in conjugation. The pattern of mt I cells, consisting of a quite homogeneous lectin-binding site density spread on the cell surface, was also common to nonmating-competent cells and to immature cells. These findings suggest that in P. primaurelia pair formation involves both NeuAc and GlcNAc residues and that the development of mating-competence is related to a modification in NeuAc and GlcNAc relative distribution on the cell surface of mt 11 cells.  相似文献

Polymorphism of four enzymatic loci has been examined in 27 populations of Tetranychus urticae in relation to their geographical distribution and to two ecological parameters: open field vs. greenhouse habitats, and species of the colonized host plant. Genetic differentiation was significantly correlated to geographical distance in both types of habitat. Mite density and distribution of infested plants appear to be important factors for the population structure of T. urticae . In open field, T. urticae specimens from citrus trees were genetically more similar to other 'citrus' samples collected in different localities than they were from mites collected in the same locality on other plant species.  相似文献

SYNOPSIS. An electron microscope study of Plasmodium coatneyi in the rhesus monkey supplied information on the fine structure of trophozoites, gametocytes and of the host cell. The trophozoites resemble other mammalian malaria parasites. They do not have typical protozoan mitochondria, but instead a concentric double-membraned organelle, which, it is assumed, performs mitochondrial functions. They feed on the host cell by pinocytosis, engulfing droplets of erythrocytes thru invaginations of the plasma membranes at any region of the cell or thru the cytostome. Digestion of hemoglobin takes place in small vesicles pinched off from the food vacuole proper. Gametocytes can be clearly distinguished into macro- and microgametocytes. Macrogametocytes are covered by 2 plasma membranes, the inner one appearing thicker in some places. The cytoplasm is filled with Palade's particles and has numerous vesicles of endoplasmic reticulum and toxonemes. In microgametocytes most of the inner membrane is thickened, the cytoplasm has few Palade's particles and vesicles of the endoplasmic reticulum and does not have toxonemes. Erythrocytes with trophozoites are irregularly scallop-shaped and have elevated points with knob-like protrusions covered by a double membrane. If these protrusions are sticky they might be in part responsible for clumping and arresting the schizonts and segmenters in the capillaries. The host cell contains numerous Maurer's clefts which in some instances are continuous with the membranes of the parasite suggesting that they might originate from them.  相似文献
